The purpose of the present invention is to provide: easy and convenient preparation of a polypeptide having a structure which is not easily destroyed by an enzyme; a peptide library that can be used in preparation of such a polypeptide; and a method for preparing the peptide library. Provided is a preparation method that is for a library of disulfide-rich peptides having a structure similar to any polypeptide selected from the group consisting of conotoxins and analogs thereof, and having the ability to bind to a desired target protein, the method comprising a preparation step for preparing a cross-linked protein having a specific structure in a solution containing 10-90% of an organic solvent, by causing a protein having a desired sequence to undergo a crosslinking reaction using a crosslinking agent.
