The present invention relates to a composition for preventing, treating, or alleviating graft-versus-host disease, comprising monoclonal stem cells obtained by subfractionation culture of stem cells, and to a graft-versus-host disease treatment method using same. According to a method of subfractionation culture and propagation of stem cells of the present invention, it is possible to obtain a large quantity of desired monoclonal stem cells in a short time by rapid propagation of monoclonal stem cells, and the monoclonal stem cells obtained thereby are stem cells with enhanced therapeutic effects on graft-versus-host disease, and thus can be beneficially used as graft-versus-host disease therapeutics.
