The present invention provides an amine-coated magnetic nanoparticle for removing insoluble aggregates for separating plasmid DNA in cells and a plasmid DNA separation method using the same. The amine-coated magnetic nanoparticles of the present invention have the advantage of being compatible with the buffer used in the conventional plasmid DNA separation process, and are added to the buffer of the plasmid DNA separation process and are insoluble using a magnetic field rather than centrifugation or filtration. Removing aggregates can simplify the plasmid DNA separation process and improve efficiency.
