Anti-CD73 antibodies or fragments thereof are provided. Therefore, the antibody or fragment is the VH CDR1 of SEQ ID NO: 1, VH CDR2 of SEQ ID NO: 2, VH CDR3 of SEQ ID NO: 3, VL CDR1 of SEQ ID NO: 4, VL CDR2 of SEQ ID NO: 5, and SEQ ID NO: 6 VL CDR3, or a variant of each of these. More generally, an antibody or fragment thereof with specificity for one or more amino acid residues selected from the C-terminal half of the human CD73 protein, such as the C-terminal domain, is described. Specific epitope amino acids in these domains include Y345, D399, E400, R401 and R480. Also provided are methods of using antibodies or fragments thereof to treat and diagnose diseases such as cancer.
