The present invention relates to IgE EMPD peptide immunogen constructs and preparations thereof for the treatment of IgE-mediated allergic diseases. IgE EMPD peptide immunogen constructs have a B cell epitope peptide of more than 20 amino acids, preferably cyclic, linked via selective spacers to heterologous helper T cell (Th) epitopes derived from pathogen proteins. These peptide immunogen constructs and preparations thereof can stimulate the production of highly specific antibodies directed against the IgE EMPD peptide in vaccinated hosts and are cross-reactive with membrane-bound IgE on B lymphocytes for which IgE secretion is determined. Antibodies induced by these peptide immunogen constructs and their preparations in vaccinated hosts induce apoptosis of IgE-expressing B cells and mediate antibody-dependent cytotoxicity (ADCC), resulting in antigen-resistance in the vaccinated host. By reducing specific IgE and total IgE levels, IgE-mediated allergic pathologies can be effectively treated.
