The present invention relates to a multi-stimulation responsive drug delivery system, specifically, a fusion polypeptide capable of forming and disintegrating nanostructures in response to temperature and pH stimulation, and a drug delivery system prepared using the same. The present invention prepares a fusion polypeptide in which a PEDF 34mer peptide capable of inhibiting angiogenesis, a hydrophilic elastin-based peptide, and a hydrophobic elastin-based peptide including Histidine are bonded to each other, and the fusion polypeptide has a core-shell structure under neutral conditions. It was confirmed that nanostructures could be produced, and the nanostructures collapsed under acidic conditions.
