The present invention discloses the application of the methylation site as a diagnostic marker for disease, and specifically relates to the application of the Y chromosome methylation site as a diagnostic marker for prostate cancer. More specifically, the present invention has established a method and standard for screening chromosomal methylation sites for disease, and selected 6 Y chromosomal methylation sites for the diagnosis of prostate cancer, for example, for prostate cancer. In addition, the selected Y chromosome methylation site may be applied as a marker for diagnosing prostate cancer and for early diagnosis of disease.
