The invention relates to a microfluidic device (100) and a method (600) for separating particles of a sample (10), in particular cells or nucleic acids of a biological sample (10), comprising a separation chamber (110) for separating the particles from the sample (10), the separation chamber (10) being fluidically connected to a microfluidic loop (200), the loop (200) comprising dilution fluid (210) for diluting the sample (10) and the separation chamber (110) having a particle outlet (113 ) for discharging the separated particles and a loop outlet (114) into the microfluidic loop (200) to a filter (221) of the loop (200) for filtering the dilution fluid (210) mixed with the sample (10).
