The present invention relates to a method of extracting DNA from a biological sample containing it in which a pretreatment is carried out comprising at least the following steps of: a) mechanical lysis of the biological sample, preferably by grinding, and b) protein denaturation, preferably without proteinase, and c) bringing the sample into contact with at least one glycosidase enzyme, preferably a plurality of different glycosidases, more preferably endoglycosidases.
