Final Report on the Developmental Toxicity of 1,4-Butanediol (CAS No. 110-63-4)in Swiss (CD-1 (Trade Name)) Mice on Gestational Days 6 through 15. Laboratory Supplement. Sections 1-7

The National Toxicology Program has selected 1,4-butanediol for the evaluation ofdevelopmental toxicity in laboratory animals due to its widespread use in chemical manufacturing and its potential pharmaceutical applications. 1,4-Butanediol will be administered by gavage to Swiss albino (CD-1) mice during major organogenesis (gestational days (gd) 6 through 15). Dams will be monitored in life for clinical condition, body weight, food and water intake, and weight gain. Necropsies will be scheduled for gd 6, gd 15, and gd 17. At all scheduled necropsies, the following measures will be made: maternal body weight, liver weight, kidney weights, gravid uterine weight, number of corpora lutea, pregnancy status, and clinical condition. On gd 15 and 17, maternal liver and kidneys will be fixed in neutral buffered 10% formalin for optional histopathology, the gravid uterus will be evaluated for number and status of implantation sites and live fetuses will be weighed.
