Preclinical Toxicology Evaluation of HMBA (NSC-95580) in Beagle Dogs and Sprague-Dawley Rats. Volume 1. Beagle Dogs

Hexamethylene bisacetamide was administered to Sprague-Dawley rats by intraperitoneal or intravenous injection and to beagle dogs by intravenous infusion in a series of preclinical toxicity studies. Dogs tolerated 120 hour infusions of 40 mg/kg per hour of HMBA with little evidence of toxicity. One dog in this group had a single convulsion approximately 24 hours after its infusion was completed. The total dose of HMBA administered to this group was 4800 mg/kg (96,000 mg/sq m). The central nervous system (CNS) was the major target organ of toxicity in dogs with convulsions generally noted in dogs that received doses of 60 mg/kg per hour or higher.
