The three-dimensional (3D) reconstruction of object structures, whose optical-microscope image is degraded by diffraction effects, using the microscope optical transfer function (OTF) for the case of incoherent illumunation is described. The results show that the 3D reconstruction method improves the structural resolution of the microscope by a factor 1.5 parallel to the optical axis, and by a factor 1.8 perpendicular to the optical axis. Inverse Fourier transformation of the OTF allows the closed representation of the incoherent inverse point spread function with which a direct development in space can be performed, presenting an improvement compared to the development in frequency space. Proposals for hardware developments with a view to on-line 3D reconstruction are given.
