The structure of the plasmalemma of isolated protoplasts from stem callus cultures ofSkimmia japonicawas investigated during cell wall regeneration by means of freeze-etching and deep-etching. Three of the four theoretically possible views of the membrane—the two corresponding inner fracture faces and the outer surface—were made visible. In all cases structural alterations were found in the arrangement of the particles, which are probably responsible for the synthesis and deposit of the cellulose microfibr
