High performance liquid chromatography (HPLC), using a photodiode array detector, was applied to the practical method to analyze acute poisoning drugs and toxic compounds in serum. Drugs, such as acetaminophen, bromvarerylurea, methomyl, DEP (O, O-dimethyl 2, 2, 2-trichloro-1-hydroxyethylphosphonate), and NAC (1-naphthyl N-methylcarbamate) often found in serum in the cases of poisonig, were successfully determined by means of this HPLC system. As for the sample preparation method for serum specimens, the liquid-liquid extraction method (using an Extrelut column), the solid phase extraction method (using a Sep-Pak column), and the deproteinization method (using acetonitrile) were compared. It was found that the liquid-liquid extraction method (using an Extrelut column) provided the best chromatograms, with minimum interference by the peaks of coexistent materials often seen in the analysis of serum specimens, and that it ensured the highest yield. This report also refers to some applications to actual cases of acute poisoning by cold medicine. The HPLC method using photodiode array detector, proposed in this report, is sure to be effectively and widely utilized for analysis of poisoning and/or toxic compounds in serum, mainly because of ease and simplicity of operation procedures, including preparation of the mobile phase and extraction of target compounds.
