摘要

A protocol for the detection of AFLPs using digoxigenin labelling is presented. The protocol was tested on DNA samples from Senecio jacobaea, Echium vulgare and Hippophae rhamnoides using 2 sets of selective primers, and compared to standard radioactive labelling. Sensitivity and reliability of the digoxigenin labelled primers in the AFLP technique was of the same order as that of the radioactive assay. The percentage of polymorphic bands was much higher in the 2 monocarpic perennial species (S. jacobaea and E. vulgare) than in the woody perennial H. rhamnoides. The method is reliable, cheap, avoids the use of fluorescent or radioactive labels, and no major adjustments of current standard AFLP protocols are necessary to use the digoxigenin labelled primers.
