Passive cutaneous anaphylaxis (PCA) was provoked in BALB/c mice by either mouse IgG or rabbit IgG antibodies, or by rabbit F(ab′)2 antibody fragments using varying latent periods. Mice were pretreated with BOL-148 and pyrilamine maleate, specific antagonists for serotonin and histamine, respectively, in order to study the role of these basic amines in the PCA reactions. At least three types of PCA reactions were observed. The mouse IgG-provoked PCA was partially inhibited by each of the antagonists and totally inhibited by use of the antagonists in combination. When animals were sensitized by rabbit IgG using a 1- hour latent period, the reaction was found to involve histamine, but not serotonin. The reactions provoked by either rabbit IgG, using a 3- hour latent period, or by rabbit F(ab’)2 antibody fragments were unaffected by either of the antagonists.
