Gingival tissue from adult patients undergoing periodontal surgery was used forin vitrocultivation. The explants were grown in three different media utilizing both tissue culture and organ culture techniques. Specimens were removed daily and processed for light and electron microscopic observation. The explants exhibited similar patterns with both culture techniques. Changes in the superficial epithelial layers were observed as early as 2 days after cultivation while the deeper spinous and basal cells maintained their integrity for as long as 5 days. Epithelial migration along the cut surface of the connective tissue resulted In complete encirclement of the explant followed by necrotic changes in the connective tissue.
