摘要

With an aim of improving the hydrolysis of Ca-caseinate in a practice, the immobilization method of trypsin tailored to the tryptic hydrolysis to obtain the caseinophosphopeptide was investigated, Covalent binding method with chitosan as a carrier was found to be superior to the physical adsorption method in terms of enzyme loading, relative activity of enzyme remained and reusability of enzyme, A packed-bed reactor, 0,01m in inside diameter, filled with immobilized trypsin, 0,08m in height, was devised to investigate the optimum operational conditions for hydrolysis of Ca-caseinate by employing the Response Surface Method, The optimum operational conditions searched were X-1 (recycle ratio) = 0.24, X-2 (substrate concentration) 2.28 (w/v), X-3 (pH) = 8.28 and X-4 (temperature) 37.7 degrees C. Under these conditions, the degree of hydrolysis (D.H,) was calculated to be 9.66 which is close to the theoretical value, 10.6.
