The conditions and requirements of an in vitro protein-synthesizing system in the embryos ofPinus thunbergiiseeds were studied. Even in the dry seed embryos, the ribosomes retained their synthetic capacity. Even after imbibition in the dark, the ribosomes did not show an increase in the activity of protein synthesis. An increase in the activity during dark imbibition was found in the 100,000×gsupernatant fraction. The activities of the cell-free systems prepared from both embryos of dark-imbibed and dry seeds were dependent on the addition of poly U. This suggests the lack or inactivity of messenger RNA in these seed embryos
