Marked changes in RNA- and DNA-synthesizing activities of the chromatin fraction were found to be connected with the conjugation process ofSpirogyrasp.RNA-synthesizing activity of the chromatin fraction was much lower in conjugating and zygote cells than in vegetatively growing cells. No significant differences were observed in the template activities of purified DNA prepared from vegetatively growing, conjugating, and zygote cells. The activities of DNA purified from vegetatively growing and zygote cells and calf thymus were strongly inhibited at different levels by binding with histones prepared from vegetatively growing and zygote cells. DNA-synthesizing activity of the chromatin fraction of conjugating or zygote cells was much lower than that of vegetatively growing cells.
