The hydrophobic packing for preparative low pressure hydrophobic interaction chromatography (HIC) on the new granulated cellulose matrix Granocel were prepared by alkylation with phenyl glycidyl ether in the presence of sodium hydroxide. The ligand concentration up to 600 #x3BC;mol/g was achieved with high reproducibility. The relative amount of eluted model protein at the fixed set of conditions was used as a criterion for the evaluation of the relative hydrophobicity of adsorbents. The relative hydrophobicity was found to depend on accessible, rather than total, amount of coupled ligand as well as on the nature of matrix.
