All rights reserved.DownloadedFromIP-12In this study we reported the development of an extensively characterized (end point assays) insulin-resistant in vitro model and comparison of anti-diabetic, anti-oxidant and anti-lipogenesis activity of Holarrhena antidysenterica (HA) leaf and bark, each extracted with 7 different solvents using the in vitro model. An in vitro model was developed by giving hyperglycaemic shock with 50 mM glucose to HepG2 cells which were initially adapted to normo-glucose condition of 5.5 mM. Diabetic characteristics were confirmed by insulin resistance, reactive oxygen species (ROS) production and lipogenesis. Leaf and bark of HA were collected from Western Ghats of Karnataka, throughout the year to avoid the seasonal variation in the phytochemicals. Samples were extracted with 8 solvents at two different temperatures. Extracts at sub-cytotoxic concentrations were compared on the model to check its effect on insulin resistance, ROS production and lipogenesis. The in vitro model showed insulin resistance setting at 50 mM glucose with increased ROS production and lipogenesis. Water extract of both leaf and bark reversed insulin resistance whereas acetone extract of bark reversed ROS production and lipogenesis. Activity guided fractionation needs to be done to arrive at individual or group of compounds from water and acetone extract.
