Abstract.An automated complement fixation test using a continuous flow system based on AutoAnalyzer components in which sheep cell hemolysis is detected in an optical cell counter has been developed. With this system 1,680 donor bloods diluted 1/10 were tested in parallel with manual immunodiffusion (ID) and microtiter complement fixation (CF) tests; 16 positive samples were found. The manual and automated CF tests were in agreement in 14 of these but in 2 additional instances, where the automated test was negative, the manual test gave a minimal positive titer of 8. With ID, 9 of the 16 were positive. The automated test results are available in 45 min at a rate of 60/h. The system is mechanically compatible with other automated blood banking systems.
