Somatic embryogenesis and plantlet formation were achieved from leaf and leaf-derived callus culture of Sapindus mukorossi Gaertn. Callus subcultured on MS medium containing lower levels of kinetin (0.2-0.5 mg l(-1)) and higher levels of potassium nitrate (2900 mg l(-1)) resulted in embryogenic callus often with globular embryos. Rapid and enhanced rate of embryo formation was achieved on transfer of embryogenic callus to an embryo proliferation medium containing kinetin (0.5 mg l(-1)) and a different source of nitrogen with or without gibberellic acid (0.2 mg l(-1)). Increased level of gibberellic acid restricted embryo multiplication and led to embryo maturation. Use of kinetin (2.0 mg l(-1)) along with ammonium chloride (50 mg l(-1)) and coconut milk, 10% (v/v) in the medium proved to be effective for embryo germination and plantlet formation.
