Preparation of glycopolymer micelle for application in drug and gene delivery

We developed a novel gene and drug delivery system consisting of curdlan backbone and poly(2-dimethylaminoethyl methacrylate) (PDMAEMA) side chains. Two kinds of comb-shaped glycopolymers curdlan-g-PDMAEMAs (Cur-PDMAs) were synthesized via atomic transfer radical polymerization (ATRP), which can form micelles with particle size of 80-110 nm. Cur-PDMAs exhibited much lower cytotoxicity and better gene transfection yield than polyethylenimine (PEI). Moreover, curcumin can be released from Cur-PDMAs micelles at a steady rate with no significant burst in 0-24 h. Cur-PDMAs increased cytotoxicity of the curcumin against the cancer cells. We hope this system can find interesting medical applications, especially in the cancer treatment through a combined action of chemotherapy and gene delivery. (C) 2019 Published by Elsevier B.V.
