摘要

The HMW-GSIDx5 gene(400 bp) of Wheat Gang 82-122 was cloned by micro-dissection and PCR amplification. This fragment was used as a probe to study temporal and tissue-specific expression of HMW-GS1Dx5 gene during wheat germination by RNA dot and Northern blot analysis. The results showed that HMW-GS1Dx5 gene was Expressed from anthesis to grain filing stage, but no expression was found in leaf. The Expression level of HMW-GS1Dx5 increased from anthesis to wax ripening stage and reached the highest in wax ripening stage. HMW-GS1Dx5 mRNA level in 15 days post anthesis stage is 28 /100 of it in wax ripening stage, 40 /100 in grouting stage, 72/100 in milk Ripening stage and 54% in full ripening stage, respectively.
