Littleisknownaboutthegeneticcontrolofpreimplantationdevelopment.Wehaveisolated,characterized,andmutatedapreviouslyundescribedmousegene,emTraube(Trb),/emessentialforpreimplantationdevelopment.Similarproteincodingsequencesarefoundinrats,humans,andyeast.TheTRBproteincontainedtwoamino-terminalacidicdomains,aleucinezipper,andthreeputativenuclearlocalizationsignals.TheemTrb/emgenewasexpressedatlowlevelsubiquitouslyearlyindevelopmentandbecamerestrictedtotheliverandthecentralnervoussystemfromE11.5onward.Myc-taggedTRBproteinwaslocalizedtothenucleus,andinalargeproportionofthecellstothenucleoli.TheemTrb/emmutantembryoshaltedindevelopmentatthecompactedmorulastageatE2.5.AtE3.5theystartedtodecompactandadaylatertheydisintegratedanddied.Theobserveddefectwascellautonomous,asmutantcellsfailedtoparticipateintheformationofchimericembryos.TheemTrb/emmutantembryosshoweda50%reductionofthetotalcellnumber.Themutantembryosexhibitedapaucityofribosomes,polyribosomes,androughendoplasmicreticulum.ThispaucityofribosomestogetherwiththelocalizationofTRBtothenucleoli,thesiteofribosomesynthesis,suggeststhatTRBisinvolvedinthesynthesisofribosomes./p/div
