Previousworkhasshownthatcellsdevelopingathighdensityreleasealow-molecular-weightfactorthatcaninduceisolatedDictyosteliumdiscoideumamoebaeofstrainV12M2todifferentiateintostalkcellsinthepresenceofcyclicAMP.Wenowshowthatthisdifferentiation-inducingfactor,calledDIF,canbeextractedfromcellsduringnormaldevelopmentandthatitsproductionisstronglydevelopmentallyregulated.DIFisnotdetectableinvegetativecellsbutrisesdramaticallyafteraggregationtoreachapeakduringslugmigration.DIFlevelsareverylowintwomutantsdefectiveinaggregation.ThepostaggregativesynthesisofDIFisstimulatedbytheadditionofextracellularcyclicAMP.WeproposethatDIFisamorphogencontrollingprestalkcelldifferentiation.
