Theamountofhexosaminesandacidmucopolysaccharidespresentintheratsecondarypalateincreasesduringthecriticalstagesofpalatogenesis,namely,rotationandfusion.Thesynthesisofacidmucopolysaccharidesinvivoandinvitrointhepalatewasdeterminedbytheincorporationof3H-glucosamineandNa2S35O4.ThelabeledmucopolysaccharideswereisolatedbyDEAE-cellulosechromatographyandwereidentifiedonthebasisofseveralcriteriaashyaluronicacidandsulfatedacidmucopolysaccharides.Hyaluronicacidaccountedforapproximately60%ofthetotalacidmucopolysaccharidessynthesizedinthepalatebothinvivoandinvitro.DON(6-diazo-5-oxonorleucine),aknowninhibitorofacidmucopolysaccharidesynthesis,inhibitedtheincorporationof3H-glucosamineandNa2S35O4bypalatalshelvesinvitroby70%.
