TheanteriorandintermediatelobesofthepituitaryglandareformedfromRathkersquo;spouch.FGF,BMPandWNTsignalsemanatingfromtheventraldiencephaloninfluencepouchgrowthanddevelopment.InordertoexaminetheroleofcanonicalWNTsignalingduringpituitarydevelopmentweexaminedthepituitaryexpressionoftheTCF/LEFfamilyoftranscriptionfactors,whichmediateWNTsignalingthroughthebindingofβ-catenin.Wereportheretheexpressionofseveralmembersofthisfamilyduringpituitarydevelopmentandthefunctionalroleofonemember,TCF4(TCF7L2),intheinductionofthepituitaryprimordium.TCF4isexpressedintheventraldiencephalonearlyinpituitarydevelopment,rostraltoadomainofBMPandFGFexpression.emTcf4/emdeficientmiceexpressemFgf10/emandemBmp4/em;however,theemBmp/emandemFgf/emexpressiondomainsareexpandedrostrally.Asaresult,additionalpituitaryprogenitorcellsarerecruitedintoRathkersquo;spouchinemTcf4/emmutants.MutantsalsoexhibitanexpansionoftheemSix6/emexpressiondomainwithinRathkersquo;spouch,whichmayincreasethenumberofproliferatingpouchcells,resultinginagreatlyenlargedanteriorpituitarygland.ThissuggeststhatTCF4negativelyregulatespituitarygrowththroughtwomechanisms.ThefirstmechanismistorestrictthedomainsofBMPandFGFsignalingintheventraldiencephalon,andthesecondmechanismistherestrictionofemSix6/emwithinRathkersquo;spouch.Thus,TCF4isnecessarybothintrinsicallyandextrinsicallytoRathkersquo;spouchtoensurethepropergrowthofthepituitarygland./p/div
