Ourunderstandingofthemolecularmechanismsthatoperateduringdifferentiationofmitoticallydividingspermatogoniacellsintospermatocyteslagswaybehindwhatisknownaboutotherdifferentiatingsystems.Giventheevolutionaryconservationofthemeioticprocess,wescreenedformouseproteinsthatcouldspecificallyactivateearlymeioticpromotersinemSaccharomycescerevisiae/emyeastcells,whenfusedtotheGal4activationdomain(Gal4AD).OurscreenyieldedtheemAym1/emgenethatencodesashortpeptideof45aminoacids.WeshowthataGal4ADndash;AYM1fusionproteinactivatesexpressionofreportergenesthroughthepromotersoftheearlymeiosis-specificgenesemIME2/emandemHOP1/em,andthatthisactivationisdependentontheDNA-bindingproteinUme6.emAym1/emistranscribedpredominantlyinmouseprimaryspermatocytesandingonadsoffemaleembryosundergoingthecorrespondingmeioticdivisions.Aym1immunolocalizedtonucleiofprimaryspermatocytesandoocytesandtospecifictypeAspermatogoniacells,suggestingitmightplayaroleintheprocessesleadingtomeioticcompetence.ThepotentialfunctionalrelationshipbetweenAYM1andyeastproteinsthatregulateexpressionofearlymeioticgenesisdiscussed./p/div
