Replication-dependenthistonegenesinthemouseandemXenopus/emshareacommonregulatoryelementwithintheprotein-encodingsequencecalledtheCRASαelement(codingregionactivatingsequenceα)whichhasbeenshowntomediatenormalexpressioneminvivo/emandtointeractwithnuclearfactorseminvitro/eminacellcycle-dependentmanner.Thusfar,theαelementhasonlybeenstudiedinrodentcellsinculture,anditseffectonhistonegeneexpressionduringdevelopmenthasnotbeendetermined.HereweexaminetheroleoftheαelementinhistonegeneexpressionduringemXenopus/emdevelopmentwhichfeaturesaswitchinhistonegeneexpressionfromareplication-independentmodeinoocytestoareplication-dependentmodeinembryosaftermidblastulastage.emInvivo/emexpressionexperimentsinvolvingwild-typeorα-mutantmouseH3.2genesshowthatmutationoftheCRASαelementresultsinafourfolddeclineofexpressioninembryos,butdoesnotaffectexpressioninoocytes.Twodistinctαsequence-specificbindingactivitiesweredetectedinbothoocyteandembryonicextracts.AslowlymigratingDNA-bindingcomplexwaspresentatrelativelyconstantlevelsthroughoutdevelopmentfromtheearlieststagesofoogenesisthroughlarvalstages.Incontrast,levelsofarapidlymigratingcomplexwerehighinstageIandIIoocytes,declinedinstageIIndash;VIoocytes,remainedlowinunfertilizedeggsandcleavagestageembryos,androsedramaticallyafterthemidblastulatransition.Themolecularmassesofthefactorsformingtheslowandrapidlymigratingcomplexeswereestimatedtobeapproximately110and85kDa,respectively.DNA-bindingactivityofthe85kDaα-bindingfactorwasaffectedbyphosphorylation,bindingwithhigheraffinityinthedephosphorylatedstate.TheabruptincreaseinDNA-bindingactivityofthe85-kDaα-bindingfactoratlateblastulacoincideswiththeswitchtothereplication-dependentmodeofhistonegeneexpression.WeproposethattheconservedαelementpresentinthecodingsequenceofmouseandemXenopus/emcorehistonegenesisrequiredfornormalreplication-dependenthistoneexpressioninthedevelopingemXenopus/emembryo./p/div
