Perturbingthechangesinproteinphosphorylationaccompanyingthefirstcleavagecaninhibittheappearanceofasetofproteinswhosesynthesisisinhibitedby?±-amanitin(transcription-requiringproteins,TRPs)(W.T.PoueymirouandR.M.Schultz,1987,Dev.Biol.121,489a€“498);synthesisoftheTRPsislikelytorepresentactivationoftranscriptionoftheembryonicgenomethatoccursatthe2-cellstageduringmousedevelopment.Inthepresentstudy,wereporttheeffectsofthreedifferentinhibitorsofthecAMP-dependentproteinkinase,N-[2-(methylamino)ethyl]-5-isoquinoline-sulfonamide(H8),(Rp)-cAMPS,andproteinkinaseinhibitor(PKI),eachofwhichinhibitsthekinasebyadifferentmechanism,oncleavageof2-cellembryosandsynthesisoftheTRPs.Two-cellembryospossessPK-Aactivity,whichisinhibitedbyeachoftheseinhibitors.BothH8and(Rp)-cAMPSinhibitcleavageof2-cellembryosinaconcentration-dependentmanner;similarconcentrationsofH7,whichisalesspotentinhibitorofPK-A,donotinhibitcleavage.H8and(Rp)-cAMPSinhibitinaconcentration-dependentmannerTRPsynthesis,whereashigherconcentrationsofH7arerequiredtoinhibitTRPsynthesis.MicroinjectedPKIalsoinhibitssynthesisoftheTRPs.Inaddition,H8inhibitstheaccumulationoftranslatablemessengerRNAsthatarelikelytoencodefortheTRPs.Last,H8,butnotH7,inhibitsthephosphorylationofaphosphoproteinin2-cellembryos.ResultsofthesestudiessuggestaroleforproteinphosphorylationcatalyzedbycAMP-dependentproteinkinaseinregulatingtranscriptionintheearlymouseembryo.
