ThetransitionoftypeIandtypeIIcollagensduringcartilageandbonedevelopmentinthechickembryowasstudiedbyimmunofluorescenceusingantibodiesagainsttypeIortypeIIcollagens.TypeIIcollagenwasfoundinallcartilaginousstructureswhichshowedmetachromaticstaining.TypeIcollagenappearedintheperichondriumofthetibiaatstage28andwasalsofoundinosteoid,periostealandenchondralboneafterdecalcification,periosteum,andtendons,ligaments,andcapsules.Usingtheimmunohistologicalmethoditwaspossibletoidentifyspecificcollagentypesinareasundergoingrapidproliferationandcollagentransition,suchasdiaphysealandepiphysealperichondrium,orinenchondralosteogenesis.DuringenchondralossificationtypeIcollagenisdepositedontotheerodedsurfaceofcartilage.Itpartiallydiffusesintothecartilagematrixformingaa€?hybrida€?collagenmatrixwithtypeIIcollagen,whichisasiteforsubsequentossification.Duringappositionalgrowthofdiaphysealcartilageanddifferentiationofepiphysealperichondriumintoarticularcartilage,perichondralcellsswitchfromtypeItotypeIIcollagensynthesiswhendifferentiatingintochondroblasts.Inthetransitionzones,chondroblastsareimbeddedinaa€?hybrida€?matrixconsistingofamixtureoftypeIandtypeIIcollagens.
