MutationsintheNF1genemaycausedevelopmentalabnormalitiesandtheformationofavarietyoftumorsofneuralcrestorigininhumans.TheNF1genecodesforalargeprotein,neurofibromin(nf),whichisstructurallyandfunctionallyrelatedtoyeastandhumanras-GTPase-activatingproteins(ras-GAPs).Recently,twotranscriptscodingfortypeIandtypeIInfwithdifferentras-GAPactivityhavebeenidentified.Sincerasproteinsdonotappeartobesignificantlyregulatedduringmousedevelopment,weexaminedifdifferentialexpressionofneurofibrominsmayprovideevidenceforaroleofnfsinregulatingras-mediatedcellproliferationanddifferentiation.NfswereexpressedasearlyasE8.AtE11amarkedincreaseofNF1transcriptsoccurredandwasassociatedwithexpressionofnfsinalltissues.TypeIandtypeIInfseachshowedadifferenttimecourseofexpressionandtissuelocalization,withtypeIInfpresentmainlyfromE8throughE10,althoughinthehearttypeIInfwaspresentatE12.Insometissuessuchasheartanddorsalrootgangliarapidincreasesanddecreasesofnfsweredetectedrelatedtodifferentiationofthesetissues.Theseresultsareconsistentwitharoleofnfsinregulatingras-mediatedcellproliferationanddifferentiationduringdevelopmentandsupportdistinctfunctionalrolesfortypeIandtypeIInfs./p/div
