Thereconstructionofhumanepidermisduringhealingofhumanskinwoundedaftergraftingontothenudemousewasdescribedinapreviouspaper(M.D??marchez,P.Sengel,andM.Pruni??ras,1986,Dev.Biol.113,90a€“96).Theregenerationoftheepidermalbasementmembranezone(BMZ)andthereorganizationoftheconnectivetissuearethesubjectsofthepresentstudy.Theywereinvestigatedbytwocomplementarymethods:(1)electronmicroscopytoanalyzetheBMZreorganization,and(2)indirectimmunofluorescencewithspecies-specificandcross-reactingantibodiesdirectedagainstlaminin,bullouspemphigoidantigen,mouseorhumancollagensoftypesIorIV,humanelasticfibers,fibronectin,fibrin,actin,andhumanvimentin,toexaminethespeciesoriginanddistributionofBMZandconnectivetissuecomponentsduringtheregenerationprocess.ItisreportedthatgraftedhumanskinpreservesitsownimmunologicalmarkersnotonlyintheepidermisbutalsointheBMZanddermisaswell,andthat,afterinjury,itsregenerationproceedsaccordingtothefollowingsequenceofoverlappingevents:(1)productionofamousegranulationtissue;(2)reepidermizationbyhumancells;(3)reconstructionofaBMZwithhumancharacteristics;(4)formationofahumanneodermis.Itisconcludedthathumanskingraftedontothenudemouseisabletoregenerateitsthreestructuralcompartments,namely,theepidermis,BMZ,anddermis.Interestingly,itappeared,also,thattheconnectivetissueregenerationwouldbeatwo-stepmechanismincludingthesequentialformationoftwotissuesofdistinctsources,namely,agranulationtissueandaneodermis.
