ThePhillymousedevelopsahereditarycataractabout5weeksafterbirth.Althoughthecausativeagentisnotknown,datasuggestthatthereisacorrelationbetweencataractformationandtheselectiveabsenceofa27kilodalton(27K)?2-crystallinlenspolypeptide.Theontogenyofthe27K?2-crystallinpolypeptidewasexaminedinnormalmiceinordertoevaluateitsroleinnormaldevelopmentanddeterminewhatimpactitsabsencemayhaveonthePhillymouselens.AmonoclonalantibodywasusedwiththePAPmethodtoimmunocytochemicallylocalizethe27Kpolypeptideinlensesofnormalmiceduringdevelopment.?2-Crystallinsdetectedwithpolyclonalantiserawerefoundindifferentiatedfibercellsthroughoutthelens.Incontrast,the27K?2-crystallinpolypeptidedetectedwithaspecificmonoclonalantibodywasnotfoundinthefibercellsoftheinnerpartofthelens(nucleus),butwasspecificallylocalizedinthefibercellsoftheouterpartofthelenscalledthecortex.Thepolypeptidewasfoundonlyinelongatinganddifferentiatedfibercellsandnotinmitoticallyactiveepithelialcells.Althoughaminorcomponentofthe2-day-oldlens,the27Kpolypeptidecomprisedalargeportionofthe16-day-oldlensincludingtheanteriorandposteriorpoles.Thesedatashowthatthe27Kpolypeptideisaminorcomponentoftheembryoniclens,butbecomesamajorcontributortothepostnatallens.The27K?2-crystallinlenspolypeptideisabundantinthefibercellsofthenormalpostnatalmouselens.Theabsenceofthe27KpolypeptideinthePhillymousemaycontributetotheobservedfailureoffibercellstodifferentiateinthePhillymouseafterbirthormaybedeleteriousinsomeothermannertonormallensdevelopment.Theselectiveabsenceofthe27K?2-crystallinpolypeptide,adefectwhichprecedescataractformationinthePhillymouse,isintriguingsinceitsuggestsarelationshipbetweenthismajorlenspolypeptideandlensclarity.
