Anti-light chain antibodies that react well with separated light chains and intact immunoglobulins of human beings could be readily produced in rabbits and goats by immunization with the Fab fragment of normal human IgG. Antibodies against light chains of κ or λ type were specifically purified from rabbit anti-normal Fab sera and rendered monospecific by the exclusive use of solid immunoadsorbents coupled with appropriate antigens. The anti-κ or anti-λ antibodies thus prepared were conjugated with fluorescein and tetramethylrhodamine and used for the study of immunoglobulin production in individual cells of hematopoietic origin in established culture.
