VP64iswidelyusedasatranscriptionalactivatortoinvestigatethebiologicalfunctionofgenes,butitspotentialforapplicationingeneticimprovementofcropshasnotbeenfullyinvestigated.Here,wecharacterizedanOsMYB1R1–VP64fusionproteinthatenhancedthegrainyieldofricecultivar‘Kita‐ake’by35%.OsMYB1R1–VP64regulatedgrainyieldoftransgenicplantsmainlybyextendingthevegetativegrowthstage.FurtheranalysisindicatedthatOsMYB1R1–VP64delayedfloweringbydown‐regulatingexpressionofHeadingdate3a(Hd3a)andRICEFLOWERINGLOCUST1(RFT1)throughrepressionofEarlyheadingdate1(Ehd1).TheexpressionpatternofOsMYB1R1wasconstitutiveandrhythmicallycontrolled.OsMYB1R1proteinhadtranscriptionalactivationactivityandwaslocalizedtothenucleus.OurfindingssuggestthattheOsMYB1R1–VP64constructcanbeusedtoincreasegrainyieldinrice...
