摘要

Cryopreservation of fish sperm is an important toolfor conservation of biodiversity (gene banks ofautochthonous populations), efficient and selectivefertilization (maintaining genetic variability of broodstocks for restocking), and for synchronization ofartificial reproduction [1]. However, cryopreservationcauses irreversible cell damage, resulting in a significantreduction in viable spermatozoa. Multiplenonphysiological processes, such as osmotic andoxidative stress, are crucial factors in spermatozoacryodamage. Freezing of cells in suspension induces icenucleation in the extracellular space, creating an osmoticgradient across the plasma membrane between theinitially isotonic intracellular solution and the freezeconcentratedextracellular solution. Hence, we designeda series of experiments using sperm of common carp(Cyprinus carpio, L.) as a model to understand theeffects of the application of different cryopreservationprotocols and to examine cryopreservation procedureswith respect to osmotic and oxidative stress in commoncarp spermatozoa. In addition, the spermatozoa motility,membrane integrity, and oxidative stress indicators aswell as response of sperm proteome were measured inunfrozen sperm and after freeze/thaw processes.
