ThisstudyhasbeenconductedtoinvestigatedifferentexplantsofplantgrowthregulatorsoncallusinductionofCalendulaofficinalisL.medicinalandaromaticherb.Theseedsweresurfacesterilizedusing70%ethanolfor3minutes,10%commercialbleachfor5minutesandrinsed3timeswithsterilewaterfor3minutes.TheseedsweregerminatedinMurashigeandSkoog(MS)medium.Hypocotyl,cotyledonandcotyledonnodeexplantshavebeenexcisedfromtheplantletsobtainedfrominvitrogerminatedseeds.ThreeexplantswereculturedonMSmediasupplementedwithvariousconcentrationsofcytokinin(BAP;1.0,2.0mgL-1)andauxin(IBA;0.1,0,5mgL-1)forcallusinduction.Attheendofeightweeks,thebestresultswereobservedinatreatmentwith1mgL-1BAP+0.5mgL-1IBAand2mgL-1BAP+0.5mgL-1IBA100%onthehypocotyl;1mgL-1BAP+0.5mgL-1IBAand2mgL-1BAP+0.5mgL-1IBA88%onthecotyledon;2mgL-1BAP+0.1mgL-1IBA100%onthecotyledonnodes.Thedataobtainedfromthepathisinstructivetogenetransformation,phytoremediationandcellculturestudy.
