RNA-seq. analysis in the amygdala revealed up- regulation of histone methylation-related genes after reactivation, suggesting a possible molecular mechanism underlying fear mem- ory persistence regulated by reactivation. By using combination of two different opsins, Chronos and ChrimsonR, we found that reactivation effect was input specific. In vivo field recording demon- strated LTP was induced at optogenetic CS (OCS) inputs after OFC training and maintained over time in a reactivation-dependent manner. These results reveal that reactivation of CS input synapses to the LA serves as a systems-level mechanism for fear mem- ory persistence. Our study provides novel insights into likely how fear memory to the specific CS is selectively maintained over time within overlapping engram cell networks.
