Membrane estrogen receptor stimulation of corticotropin-releasing hormone expression in a nerve cell line Sex-specific changes of steroid hormone metabolism in the cerebral cortex of mice after acute valproate exposure Valproic acid (VPA), a kind of histone deacetylase inhibitor, modulates gene expression by epigenetic mechanism. There is lack of study about the effect of VPA for the reproductive endocrine sys- tem of males and females. In this study, we investigated metabolic alterations using liquid chromatography-tandem mass spectrom- etry in mice brain after VPA acute treatment. at the VPA-treated (400 mg/kg in saline solution, intraperitoneally) mice, cortisol lev- els were increased and 17-estradiol levels were decreased in both sex. In addition, in the male mice, dihydrotestosterone levels were increased, and testosterone were decreased. The 4-hydroxylase activity was upregulated in the female mice, and the 5a-reductase activity was increased in the male mice. These results indicate sex specific change of steroid metabolism in the cortex by VPA acute exposure.
