Weexaminedinvitrofertilizabilityanddevelopmentof10inbredmousestrains(C57BL/6J,C57BL/10,C57BL/10.D2ewSn,C57BL/10-Thy1.1,C57BL/10.Br/Sn,C3H/He,RFM/Ms,STS/A,BALB/c-nuandC.B-17/Icr),andtheviabilityoffrozen-thawedinvitrofertilized(IVF)embryosafterembryotransfer(ET).Insevenstrains,fertilizabilitywassignificantlygreaterinmodifiedhumantubalfluid(mHTF)comparedwithmodifiedKrebs-Ringer'sbicarbonatesolution(TYHmedium).TheTYHmediumsupportedalmostnofertilizationinfourstrains.Morethan80%ofIVFembryosdevelopedtotheblastocyststageby120hinpotassium-enhancedsimplexoptimizationmedium(KSOM).ReciprocalfertilizationbetweenC57BL/6JandBALB/c-nugametesinTYHmediumyieldedpoorfertilizationofBALB/c-nuduetospermatozoaldeficiencies.IncreasedconcentrationsofbovineserumalbuminandspermatozoaduringcapacitationandPercollwashingdidnotdrasticallyaffectfertilization.ThemHTF,butnotTYHmedium,supportedBALB/c-nuspermatozoapenetrationintothezonapellucidairrespectiveofcapacitationmedia.Invitrofertilizedembryosfrozen-thawedrapidlyweretransferredtosurrogatemothersatthetwo-cellstage.Comparedwiththatofunfrozencontrols,rapidfreezinghadnosignificanteffectonfetusdevelopmentexceptinC57BL/10.D2ewSnmice.TheseresultssuggestthatmHTFmediumissuperiorwithrespecttoIVFofinbredmice,andthatKSOMadequatelysupportsinvitrofertilizedembryodevelopmentininbredmice.ThedataalsoindicatethatrapidfreezingofpronucleateembryosfollowingIVFissuitableforcryopreservationandembryobankingofinbredmiceandfortheproductionofgeneticallymodifiedmice.
