GBF1, a cellular protein which is an activator of small Arf GTPases, is an essential host factor for enterovirus replication. The role of individual Arfs in the replication process is not well understood. Activated Arfs recruit effector proteins that regulate the biochemical composition of the cell membrane. Moghimi et al. (e01629-20) discovered that all cellular Arfs are translocated to enterovirus replication organelles in a specific temporal succession. Arf1 and Arf6 were shown to be the most important for the replication. These results underscore the unique membrane metabolism of infected cells which can be targeted by host-directed antivirals.
