► First report of a rabies virus vector based on a human vaccine strain. ► Vectors include two forms: replication competent and replication constrained. ► The activity of biomarker Gluc facilitated the comparison of different vectors. ► Gluc expression in replication constrained vectors was highest. ► The addition of glycoprotein was slightly beneficial for virus recovery.
