HPLC 法测定药用菊花中10个主要化学成分的含量

         

摘要

Objective:To establish an HPLC method for simultaneous determination of 10 contents including 5 caffeoylquinic acids and 5 flavones in Chrysanthemum Flower.Methods:HPLC separation was carried out with an DiamonsilTMC18 column(4.6 mm × 250 mm,5 μm)using acetonitrile-0.05% Formic acid as mobile phase at a flow rate of 1.0 mL/min.The detection wavelength was 335 nm and the column temperature was maintained at 40 ℃.Results:Good linear relationships were observed between the masses and the peak areas of the 10 contents;The average recovery rates were between 95.6 and 100.3%,RSD was between 0.66 and 1.95%.Conclusion:The content determination method can be used as an effective method for the quality control of Chrysanthe-mum Flower,which provides the data base for the research on the relationships between different contents of Chrysanthemum Flow-er.%目的:建立 HPLC 法测定菊花药材中10个主要化学成分的含量,包括5个咖啡酰奎宁酸类成分和5个黄酮类成分。方法:采用 DiamonsilTMC18(4.6 mm ×250 mm,5μm)色谱柱,流动相为乙腈-0.05%甲酸,流速1.0 mL/min,检测波长335 nm,柱温40℃。结果:10个化合物在一定范围内与峰面积呈良好的线性关系,r 值均大于0.9995;平均加样回收率(n =6)范围为95.6%~100.3%,RSD 为0.66%~1.95%。结论:本文所建立含量测定方法可作为中药菊花的质量控制的有效方法,为明确不同品种菊花的基原关系和品种鉴定研究提供了数据基础。
