益气通脉饮对慢性心力衰竭大鼠心功能及血清TNF-α、IL-6、TGF-β1水平的影响

         

摘要

Objective:To discuss YiQi TongMai liquid in different dosages on heart function and the levels of TNF-α, IL-6 and TGF-β1 of rats with chronic heart failure (CHF). Methods:Forty rats were randomized into model control group, equivalent dosage group of herbs, large dosage group of herbs, the control group of Western medicine, and normal control group, eight rats each group. Rat model with heart failure after acute myocardial infarction was replicated by ligation of left coronary artery main stem, after successfully establishing the models, equivalent dosage group of herbs, large dosage group of herbs accepted intragastric administration of YiQi TongMai liquid in the light of 5.4 g/kg (body weight) and 16.2 g/kg (body weight) respectively;the control group of Western medicine cozaar suspension according to the standard:4.5 mg/kg (body weight), model control group and normal control group the same volumes of physiological saline, once per day, for eight weeks. Doppler echocardiogram machine was adopted to detect heart function of CHF rat, radioimmunoassay to determine the contents of TNF-α, IL-6 and TGF-β1. Re-sults:The comparison of ESV, LVEF, EDV and SV had significant difference between model control group and nor-mal control group (P<0.05), and the difference had statistical meaning;compared with model control group, there was significant difference in ESV, LVEF, EDV and SV of equivalent dosage group of herbs and the control group of Western medicine (P<0.05), and the differences had statistical meaning;while there was no significant difference in the comparison of all indexes between large dosage group of herbs and the control group of Western medicine (P>0.05), compared with normal control group, the levels of TNF-α, IL-6 and TGF-β1 increased notably in the rats of model control group (P<0.01);these indexes decreased in large dosage group of herbs, equivalent dosage group of herbs and the control group of Western medicine compared with model control group (P<0.05), while the differ-ences had no statistical meaning among three groups in the levels of TNF-α, IL-6 and TGF-β1 (P>0.05). Conclusion:YiQi TongMai liquid could relieve the symptoms of CHF rats, decrease the levels of TNF-α, IL-6 and TGF-β1, there-fore, to delay the onset and development of CHF.%目的:探讨益气通脉饮不同剂量给药对慢性心力衰竭(CHF)大鼠心功能以及血清TNF-α、IL-6、TGF-β1水平的影响。方法:将40只大鼠随机分为模型对照组、中药等效剂量组、中药大剂量组、西药对照组、正常对照组,每组8只。采用结扎大鼠左冠状动脉主干的方法复制急性心梗后心衰大鼠模型,造模成功后,中药等效剂量组与大剂量组分别按照5.4 g/kg(体质量)、16.2 g/kg(体质量)予益气通脉饮煎取液灌胃;西药对照组予科素亚混悬液,按4.5 mg/kg(体质量)灌胃,模型对照组与正常对照组以同体积生理盐水灌胃。1次/d,共8周。采用多普勒超声心动图机测定CHF大鼠的心功能,用放射免疫法检测大鼠血清中TNF-α、IL-6、TGF-β1的含量。结果:模型对照组ESV、LVEF、EDV、SV与正常对照组比较差异有统计学意义(P<0.05);与模型对照组比较,中药等效剂量组、西药对照组ESV、LVEF、EDV、SV有统计学差异(P<0.05);而中药大剂量组和西药对照组比较各项指标无明显差异(P>0.05),与正常对照组相比,模型对照组大鼠血清TNF-α、IL-6、TGF-β1水平显著增高(P<0.01);中药大剂量组、等效剂量组和西药对照组血清TNF-α、IL-6、TGF-β1水平均较模型对照组降低(P<0.05)。而中药大剂量组、等效剂量组和西药对照组之间血清TNF-α、IL-6、TGF-β1水平比较差异无统计学意义(P>0.05)。结论:益气通脉饮可改善CHF大鼠的心衰症状,降低血清中TNF-α、IL-6、TGF-β1水平,从而延缓慢性心力衰竭的发生和发展。
