To observe inducing effect of nerve regeneration on peripheral nerve defect models bridged by nerve conduct tube filled with pyrroloquinoline quinone(PQQ).Methods This study was carried out on 24 male SD rats randomly divided into pyrroloquinoline quinone group and normal saline group,12 rats in each group.Models of 1.0 cm sciatic nerve defect were created in this experiment.The nerve defect was bridged by tubes filled with pyrroloquinoline quinone(10 μL,0.03 mmol/L) in the experimental group while by same volume of normal saline in the control group.Motor nerve conductive velocity (MCV) was measured and morphology study was carried out under light microscope and electronic microscope 12 weeks after intubulation.A digital image-analysis system was used to measure the thickness of myelin sheath and count the regenerated axon number.Results MCV(m/s) is those two groups were 23.95±2.47 and 19.46±2.46 respectively;regenerated axon number,were 3060±223 and 2300±250;thickness of myelin sheath(μm) were 0.437±0.059 and 0.290±0.031.There exists a statistic difference between those two groups,and animals in PQQ group showed a better recovery.More mature nerve regeneration could also be observed in the PQQ group when morphological study was carried out.Conclusion Pyrroloquinoline quinone takes an active role in the process of peripheral nerve regeneration,and regeneration was greatly accelerated by it.7 refs,1tab.
