无患子组培过程中愈伤组织诱导和芽苗增殖的培养基配方研究

         

摘要

To explore the effect of macroelements and plant hormones on the callus inducement and shoot growth in tissue culture of Sapindus mukorossi Gaerth. , and get the optimal medium to culture the plant in vitro. , 9 different media were made with three factors, i. e. macroelements of MS, BA and NAA with three levels according to L9 (34 ) orthogonal array to culture stem segments of S. mukorossi Gaerth. Data on growth got after 30 days were analyzed by range analysis and avriance analysis. The results showed that BA was the main factor that affected the callus inducement and shoot growth, and then was macroelement of MS, and NAA had no significant effect. Full MS and low concentration of BA (2.0 mg · L ‐1) were both the best for inducing the redifferentiation of callus and spouting of bud seedling. Results also showed that there was a mutual restriction and mutual promoting relationship between the callus induction and sprout propagation. The optimized composition was medium No. 2 in this study, and the theoretical optimized one was MS + BA 2.0 mg o L ‐1+ NAA 0.01 mg o L‐1.%为探讨大量元素和植物激素对无患子愈伤组织诱导和芽苗增殖的影响并优化培养基组成,试验采用正交设计研究了不同浓度的大量元素、BA和NAA对无患子茎段培养的影响,并对试验数据进行极差分析和方差分析.结果表明BA在无患子茎段愈伤组织诱导和芽苗增殖中起主要作用,其次为MS大量元素,NAA无明显作用.全量MS和2.0 mg·L-1的BA最适于无患子茎段上愈伤组织的诱导与再分化以及芽苗的增殖和伸长;MS大量元素含量降低、BA和NAA浓度增加均不利于无患子离体培养物的生长,这在愈伤组织的再分化上表现尤为显著.无患子茎段培养的最适培养基是MS+BA2.0mg·L-1+ NAA0.01 mg·L-1,本试验中MS+BA2.0mg· L-1+ NAA0.05mg·L-1即2号培养基为最佳组合.
